Tomography and Cryo-FIB Workshop 2024

This workshop will help you learn the fundamentals of cryo-electron tomography (cryo-ET) including sample preparation, FIB-milling, correlative imaging, and cryo-ET data collection and analysis. Whether you are new to the field, have been collecting single-particle cryo-EM data and are now looking to start your first tomography project, or have experience with tomography, you'll come away with fresh ideas and strategies you can put to use right away. The morning sessions of the workshop will be hybrid and the afternoon practical sessions will be in-person only. Connect face-to-face with colleagues and build community with other tomography researchers at the Midwest Center for Cryo-Electron Tomography (MCCET) at the University of Wisconsin - Madison. 

Topics include: 

  • Introduction to cryo-ET including the basic principles of dose tolerance, illumination, and tilt series collection parameters. 
  • Training in data acquisition workflows that include screening, data collection, and flexible scripting.
  • Training in basic data processing including pre-processing, tomogram generation, and considerations for segmentation and sub-tomogram averaging. 
  • Instruction in 2D/3D correlative imaging workflows using the Leica widefield and cryo-confocal microscopes, and the integrated fluorescence microscope in the Aquilos 2. 
  • Guidance in FIB-milling with AutoTEM and advanced FIB workflows such as waffle milling and lift-out methods. 

WHEN: Monday, September 23, 2024, to Friday, September 27, 2024 
Breakfast begins at 8:00 a.m. followed by the morning program from 9 a.m. to 12:00 p.m. 
Afternoon breakout sessions for hands-on use of instrumentation until 5:00 p.m. 
Meals are included.
